Biologically Relevant Murine Models of Chronic Pseudomonas aeruginosa Respiratory Infection

چکیده
Pseudomonas aeruginosa (P. aeruginosa) is an opportunistic pathogen and the leading cause of infection in patients with cystic fibrosis (CF). The ability P. to evade host responses develop into chronic causes significant morbidity mortality. Several mouse models have been developed study respiratory infections induced by aeruginosa, bead agar model being most widely used. However, this has several limitations, including requirement for surgical procedures high mortality rates. Herein, we describe novel adapted biologically relevant lung caused aeruginosa. Three methods are described: a clinical isolate model, utilising isolates obtained from CF; incomplete antibiotic clearance bacterial bounce-back; establishment infection; water bottle model. These circumvent procedure and, importantly, can be wild-type mice. We also demonstrate successful induction transgenic βENaC murine CF. envisage that described will facilitate investigations microbial factors, efficacy antimicrobials, during infections.

In vivo efficacy of doripenem (DRPM) against Pseudomonas aeruginosa in murine chronic respiratory tract infection model.
Doripenem is a carbapenem antibiotic with broad-spectrum coverage of gram-positive and gram-negative bacteria, including Pseudomonas aeruginosa, and is considered to be as effective as meropenem. The in vivo activity of doripenem was thus compared with that of meropenem in a chronic lower respiratory P. aeruginosa infection mouse model. The number of viable bacteria in the lungs of mice after t...

Respiratory infections caused by Pseudomonas aeruginosa are a major clinical problem globally, particularly for patients with chronic pulmonary disorders, such as those with cystic fibrosis (CF), non-CF bronchiectasis (nCFB) and severe chronic obstructive pulmonary disease (COPD). In addition, critically ill and immunocompromised patients are also at significant risk of P. aeruginosa infection....

Diffuse panbronchiolitis (DPB) is a chronic lower respiratory tract infection commonly associated with persistent late-stage Pseudomonas aeruginosa infection. However, low-dose long-term therapy with certain macrolides is effective in most patients with DPB. The present study was designed to examine the effects of long-term erythromycin (ERY) therapy by using our established murine model of chr...
متن کاملMercaptoethylguanidine inhibits the inflammatory response in a murine model of chronic infection with Pseudomonas aeruginosa.
Chronic airway inflammation induced by Pseudomonas aeruginosa is the eventual cause of respiratory failure in most people affected by cystic fibrosis. Recent evidence implicates the involvement of free radical and oxidant stress in the pathogenesis of the inflammatory injury. Here we report the efficacy of a novel experimental therapeutic, mercaptoethylguanidine (MEG), which has combined action...
متن کاملEffect of anti-PcrV antibody in a murine chronic airway Pseudomonas aeruginosa infection model.
Pseudomonas aeruginosa is one of the most important pathogens in patients with chronic airway conditions, such as cystic fibrosis and diffuse panbronchiolitis. Type III secretion system-mediated virulence factors contribute to the lung damage in chronic P. aeruginosa infection. The effects of the anti-PcrV immunoglobulin (Ig)G, which blocks the type III secretion system, were evaluated in a mou...
